Purification of immunoglobulin Y from egg yolk using thermoresponsive aqueous micellar two-phase systems comprising ionic liquids

Immunoglobulin Y (IgY) represents an important class of antibodies, being present in egg yolk, with relevant medical applications and involving non-invasive methods extraction. However, due to the complexity purification levels required for use most demands application multi-step cost-intensive techniques. Therefore, it is upmost importance develop a biocompatible cost-effective downstream process purify IgY from yolk. In this work, was purified yolk water-soluble protein fraction (WSPF) by thermoresponsive aqueous micellar two-phase systems (AMTPS) composed non-ionic surfactant Triton X-114 surface-active ionic liquids (SAILs) as co-surfactants. The best allowed, single step, recover surfactant-poor phase purity level 69%, up 73% if consecutive extraction cycles are performed, while maintaining structural integrity antibodies. Considering these results, designed proposed, consisting four main steps: (i) recovery WSPF; (ii) applying AMTPS; (iii) isolation solvents using ultrafiltration step; (iv) recycling AMTPS phase-forming components precipitating contaminant proteins.
